No one knows for sure. A living human brain is extremely hard to study. More is being learned all the time.
Depression is a very broad term with many possible causes.
I believe in my case, for many reasons, that it is primarily caused by genetics and biology. Meaning changes in neurotransmitters, receptors, neuro pathways, synatpogenisis, neuro genesis, or whatever, precede the depression.
