Yep. Pharmacists know all of this information because they're legally responsible for making sure there are no drug interactions with your existing Rx's. If your doctor sends them a new Rx and it interacts with your existing Rx's, they reject the new Rx. The only exception is if your PRN's have interactions with other PRN's; they'll just warn you about the interactions before approving it.

So while they're legally not responsible for OTC med interactions with Rx meds, they'll know for sure and they HAVE to tell you if you ask.
