The affect (action) of the medication may cause drowsiness.

I have felt the effect (noun) of the medication and it made me drowsy.

Effect is a betterr word in both cases! Medication may affect (verb, action word) drowsiness. You have used affect as a noun: the "affect" of the medication...

Noun verb object...

Medication affect drowsiness...
